Purification and activation of Clostridium botulinum type E toxin.

Type E strains of Clostridium botulinum tend to produce toxin of relatively low mouse killing power (administered intraperitoneally) in relation to its high lethality per os for man. The explanation of this peculiarity seems to lie in the phenomenon of "activation," whereby type E toxin, when exposed at an appropriate pH in vitro to certain bacterial enzymes (Dolman, 1953, 1957a; Sakaguchi and Tohyama, 1955) or to trypsin (Duff, Wright, and Yarinsky, 1956; Dolman, 1957a), acquires enhanced lethality for mice. The same process of tryptic activation, occurring in vivo near the gastroduodenal junction, has been suggested as a possible explanation of the high mortality rate in some type E botulism outbreaks (Dolman, 1957b, Dolnan et al., 1960). The mechanism of this activation phenomenon is not yet clear. Sakaguchi and Sakaguchi (1959) have claimed that type E toxin is associated in some way with ribonucleic acid, and that after activation with trypsin the toxic fraction is separated from its nucleic acid component. This report outlines experimental findings at variance with that claim. In our hands, toxic preparations, subjected to partial purification before and after activation with trypsin, were free from nucleic acids.
